Formazan Formation Measurement

Formazan formation measurement is a colorimetric method that quantifies the colored products generated when tetrazolium salts are reduced, providing an indirect readout of cellular metabolic activity. In living cells, NAD(P)H-dependent oxidoreductases transfer electrons to tetrazolium compounds, producing insoluble or soluble formazan; the product’s absorbance is measured spectrophotometrically and related to reducing activity under defined conditions. In biology, assays based on MTT, XTT, or related reagents help estimate cell viability, proliferation, cytotoxicity, and metabolic responses. Because signal intensity depends on both cell number and metabolic state, experimental controls and standardized incubation conditions are essential for reliable interpretation.

This method allows selective staining and quantification of DNA in gels by soaking the gel in a SYBR Green I/Nitro Blue Tetrazolium solution and then exposing it to sunlight or a blue light source. This produces a visible precipitate and requires almost no equipment, making it ideal for field use.
