Glucose 6-phosphate

Glucose 6-phosphate is a phosphorylated form of glucose and a central metabolic intermediate that links carbohydrate breakdown, biosynthesis, and cellular energy management. Cells produce it when hexokinase or glucokinase transfers a phosphate group to glucose using ATP; the added phosphate helps retain glucose inside the cell and directs it into pathways such as glycolysis, the pentose phosphate pathway, or glycogen synthesis. Measuring glucose 6-phosphate and the enzymes that regulate its formation helps researchers assess metabolic flux, oxidative-stress defenses, and glucose homeostasis. Its roles are especially important in studies of diabetes, inherited metabolic disorders, cancer metabolism, and cellular adaptation.

This protocol uses G6PI mixed peptides to construct rheumatoid arthritis models that are closer to that of human rheumatoid arthritis in CD4+ T cells and cytokines. High purity invariant natural killer T cells (mainly iNKT2) with specific phenotypes and functions were obtained by in vivo induction and in vitro purification for adoptive immunotherapy.

Assessing Glucose Uptake in the Motor Neurons of Drosophila Larvae with Upregulated Glucose Metabolism

2025

The video demonstrates a technique for measuring glucose uptake in Drosophila larval motor neurons using a fluorescence resonance energy transfer (FRET)-based intracellular glucose sensor. After dissecting the larvae to expose the ventral nerve cord, the motor neurons are visualized through the fluorescence emitted by the sensor's donor fluorophore. When stimulated with glucose, FRET between the sensor's donor and acceptor fluorophores triggers the emission of fluorescence from the acceptor,...

This video demonstrates an in vitro assay to indirectly measure intracellular glucose uptake. The assay uses a fluorescently-labeled glucose analog to establish a correlation between extracellular glucose depletion and intracellular glucose uptake.

Measurement of Extracellular Fluorescent Glucose Analog Depletion: A Surrogate Measurement Technique to Assess Glucose Uptake in Mouse Organs Ex Vivo

2025

In this video, we demonstrate an ex vivo technique to measure extracellular glucose analog depletion in harvested mouse organs. It involves incubating the mouse tissue in a solution containing fluorescently-labeled deoxyglucose, which is taken up by the cells. The glucose analog uptake can be indirectly determined by measuring the fluorescence of the incubated medium.
