Phospholipase C

Phospholipase C is a family of enzymes that regulates cell signaling by cleaving membrane phospholipids, making it a central component of communication between cells and their environment. In a common pathway, activated G protein-coupled receptors or receptor tyrosine kinases stimulate PLC to hydrolyze phosphatidylinositol 4,5-bisphosphate into inositol trisphosphate (IP3) and diacylglycerol (DAG). IP3 promotes calcium release from the endoplasmic reticulum, while DAG remains in the membrane and helps activate protein kinase C. These coordinated signals influence secretion, contraction, metabolism, proliferation, and gene expression, making PLC important in studies of physiology, development, immunity, and disease.

In this video, we demonstrate an assay to establish the presence of phospholipase in the crude venom extract from Anthopleura dowii—a marine sea anemone. The assay utilizes an agarose gel diffusion technique, in which the enzyme phospholipase diffuses and breaks down phospholipids incorporated in the solid media in a radial manner.

Detection of Phospholipase C Activity in the Brain Homogenate from the Honeybee

To test the inhibitory effects of pharmacologic agents on phospholipase C (PLC) in different regions of the honeybee brain, we present a biochemical assay to measure PLC activity in those regions. This assay could be useful for comparing PLC activity among tissues, as well as among bees exhibiting different behaviors.

Hormones—or any molecule that binds to a receptor, known as a ligand—that are lipid-insoluble (water-soluble) are not able to diffuse across the cell membrane.
