Heart Block

Heart block is a cardiac conduction disorder in which electrical signals move slowly or fail to pass from the atria to the ventricles, disrupting the heart’s normal rhythm and pumping coordination. It usually results from impaired conduction through the atrioventricular node or the His-Purkinje system and is classified as first-, second-, or third-degree block according to the extent of signal interruption. In biology and medicine, studying heart block helps explain how specialized cardiac cells coordinate contraction and how conduction pathways fail. Electrocardiography identifies characteristic timing changes, while severe cases may require an artificial pacemaker to maintain an appropriate heart rate.

Developmental studies in the mouse are hampered by the inaccessibility of the embryo during gestation. To promote the long-term culture of the embryonic heart at late stages of gestation, we developed a protocol in which the excised heart is cultured in a semi-solid, dilute Matrigel.

The cuff technique shortens and facilitates the model of heterotopic cervical heart transplantation in mice by avoiding technically challenging suture anastomoses of small vessels. The technical details shown in this video paper should allow researches to establish this model in their laboratories.

Use of zebrafish for cardiovascular research is expanding towards research on adult hearts. For these applications, quick and simple isolation of cardiac tissues is key to avoid post-mortem changes and to obtain an adequate number of samples. Here, we describe a fast and reproducible method for dissecting adult zebrafish hearts.
