Hesc-rpe Transplantation

hESC-RPE transplantation is an experimental cell-replacement approach that delivers retinal pigment epithelial cells derived from human embryonic stem cells to restore support for damaged or lost retinal tissue. The transplanted cells are intended to integrate beneath the photoreceptor layer, where retinal pigment epithelium maintains the blood-retina barrier, supports photoreceptor metabolism, and phagocytoses shed outer segments. In biology and regenerative medicine, this strategy is being investigated for retinal degenerative disorders, including conditions involving RPE dysfunction and progressive vision loss. Research focuses on cell survival, tissue integration, immune compatibility, and whether transplantation can preserve or improve visual function.

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.
