Hit Confirmation

Hit confirmation is the process of validating candidate “hits” identified in biological or chemical screening before advancing them as credible research leads. Researchers retest each hit in replicate, measure concentration-dependent responses, and use orthogonal assays or counter-screens to determine whether the observed activity reflects the intended biological mechanism rather than assay interference, nonspecific effects, or compound toxicity. In drug discovery, hit confirmation helps distinguish reproducible, selective, and biologically meaningful activity from false positives. Reliable confirmation supports target validation, structure–activity studies, and prioritization of compounds for hit-to-lead optimization, improving the efficiency and quality of downstream research.
