Pathogen Exposure

Pathogen exposure is contact between a host and a disease-causing microorganism or agent, creating the possibility of colonization, infection, or an immune response. Exposure occurs through routes such as inhalation, ingestion, mucous-membrane contact, or breaks in the skin, while the outcome depends on pathogen dose, tissue entry, replication, and host defenses. In biology, studying pathogen exposure helps researchers trace transmission, measure susceptibility, evaluate barriers such as vaccination or protective equipment, and design safer experiments and public-health interventions across populations and settings.

In the current climate of scarce resources, new technologies are emerging that allow researchers to conduct studies cheaper, faster and with more precision. Here we describe the development of a bead-based salivary antibody multiplex immunoassay to measure human exposure to multiple environmental pathogens simultaneously.

Single Cell Measurements of Vacuolar Rupture Caused by Intracellular Pathogens

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.

A whole-body nanoparticle aerosol inhalation exposure facility was constructed for nano-sized titanium dioxide (TiO2) inhalation toxicology studies. This system provides nano-TiO2 aerosol test atmospheres that have: 1) a steady mass concentration; 2) a homogenous composition free of contaminants; and 3) a stable particle size distribution during aerosol generation.
