Imprinting

Imprinting is a form of rapid, durable learning in which an animal develops a lasting preference or recognition response toward a parent, species, habitat, or other stimulus during a sensitive early-life period. It occurs when exposure to salient sensory cues, such as visual, auditory, or olfactory signals, interacts with developmental predispositions to establish behavior with limited later modification. In biology, imprinting helps explain offspring–parent recognition, social attachment, species identification, and mate choice. Studying this process connects developmental biology with animal behavior and ecology, while informing conservation efforts involving hand-reared or relocated animals.

Imprinting is a phenomenon in plant and mammal reproduction. DNA methylation plays an important role in mechanisms of imprinting. Isolating endosperm and determining methylation status of imprinted genes in Arabidopsis can be difficult. In this protocol, we describe how to isolate endosperm and determine methylation by bisulfite sequencing.
