Intracellular Cytokine Staining

Intracellular cytokine staining is a flow cytometry technique that identifies cytokine-producing cells by detecting proteins retained inside individual cells. After cells are stimulated to promote cytokine production, secretion is blocked, and the cells are fixed and permeabilized so fluorescently labeled antibodies can enter and bind specific intracellular cytokines. Flow cytometry then measures fluorescence alongside surface markers, revealing which immune cell populations produce particular cytokines and in what proportions. This method supports analysis of cellular immune responses, functional profiling of lymphocytes, vaccine and infection studies, and evaluation of immune-modulating treatments at single-cell resolution.

Described here is a method for producing exogenous cytokine in patient-derived xenograft (PDX) mice via weekly intraperitoneal injection of a cytokine-transduced stromal cell line. This method broadens the utility of PDX and provides the option for transient or sustained exogenous cytokine delivery in a multitude of PDX models.
