Larval Tracking

Larval tracking is the systematic observation and analysis of larval movement, position, and development over time, providing a way to quantify behavior in biological systems. Researchers typically use time-lapse imaging or repeated observations to identify individual larvae, record their coordinates, and reconstruct trajectories, allowing movement patterns to be compared across environmental conditions or experimental groups. These measurements can reveal changes in swimming, dispersal, growth, and survival, supporting studies of animal development, ecology, behavior, and environmental response. Automated image analysis can further increase the scale and consistency of tracking while reducing manual scoring.

Drosophila Larval IHC

2023

Immunohistochemistry (IHC) is a technique used to visualize the presence and location of proteins within tissues. Drosophila larvae are particularly amenable to IHC because of the ease with which they can be processed for staining. Additionally, the larvae are transparent, meaning that some tissues can be visualized without the need for dissection. In IHC, proteins are ultimately detected with antibodies that specifically bind to “epitopes” within the protein of interest. In order to preserve...

This protocol analyzes navigational behavior of Drosophila larva in response to simultaneous optogenetic stimulation of its olfactory neurons. Light of 630 nm wavelength is used to activate individual olfactory neurons expressing a red-shifted channel rhodopsin. Larval movement is simultaneously tracked, digitally recorded, and analyzed using custom-written software.
