L1 Larvae

L1 larvae are the first post-embryonic larval stage in many animals, marking the transition from an embryo or newly hatched organism to active juvenile development. After hatching, L1 larvae grow and begin tissue differentiation while responding to environmental conditions such as nutrition, temperature, and population density; they later molt into the L2 stage as their body structures and developmental programs progress. In biology, L1 larvae provide a defined starting point for studying development, gene function, physiology, and environmental responses. Their synchronized growth and measurable transition between stages also support genetic screens, toxicity assays, and research on developmental regulation.

We present a novel method for microgavage of larval zebrafish utilizing standard embryo microinjection and stereomicroscopy equipment. We demonstrate that microgavage is a safe and efficient technique useful for delivering controlled amounts of diverse materials specifically into the larval zebrafish intestinal lumen.

Here we demonstrate a novel assay to study cold nociception in Drosophila larvae. This assay utilizes a custom-built Peltier probe capable of applying a focal noxious cold stimulus and results in quantifiable cold-specific behaviors. This technique will allow further cellular and molecular dissection of cold nociception.

Source: Hiroyasu, A., D et. al., Extraction of Hemocytes from Drosophila melanogaster Larvae for Microbial Infection and Analysis. J. Vis. Exp. (2018)This video demonstrates the procedure for establishing a bacterial infection in Drosophila larvae by placing them into a bacterial droplet and pricking them with a tungsten needle. The larvae are then incubated on a grooved, nutrient-rich agar plate to study host-pathogen interactions.
