Lactobacillales Order

Lactobacillales is an order of mostly Gram-positive, non-spore-forming bacteria that includes important lactic acid bacteria and matters in biology, health, and food production. These organisms obtain energy by fermenting carbohydrates, converting sugars primarily into lactic acid; acidification can inhibit competing microbes and shape their environments. Members such as Lactobacillus, Lactococcus, Streptococcus, Leuconostoc, and Pediococcus contribute to yogurt, cheese, fermented vegetables, and other foods, while related species inhabit the gastrointestinal and reproductive tracts. Studying Lactobacillales supports research on microbial ecology, probiotics, food preservation, industrial fermentation, and host-microbe interactions, although some members can also act as opportunistic pathogens.

Lentiviral Mediated Gene Silencing in Human Pseudoislet Prepared in Low Attachment Plates

A protocol to create gene modified human pseudoislets from dispersed human islet cells that are transduced by lentivirus carrying short hairpin RNA (shRNA) is presented. This protocol utilizes readily available enzyme and culture vessels, can be performed easily, and produces genetically modified human pseudoislets suitable for functional and morphological studies.
