Langendorff Perfusion

Langendorff perfusion is an ex vivo technique that maintains an isolated heart by delivering oxygenated physiological solution through the coronary circulation, enabling controlled study of cardiac function outside the body. In the standard retrograde arrangement, fluid enters the aorta under constant pressure or flow, closes the aortic valve, and passes into the coronary arteries while the heart may continue beating. Researchers use this preparation to measure contractility, heart rate, coronary flow, and responses to drugs, ischemia, and reperfusion. Because experimental conditions can be precisely controlled, Langendorff perfusion supports investigations of cardiovascular physiology, myocardial injury, and potential cardioprotective treatments.

Langendorff Retrograde Heart Perfusion: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.

The objective of this study was to establish a method for investigating cardiac dynamics using a translational animal model. The described experimental approach incorporates dual-emission optocardiography in conjunction with an electrophysiological study to assess electrical activity in an isolated, intact porcine heart model.

This article demonstrates the feasibility of achieving longer perfusion times (4 h) of murine cardiac grafts without function loss by employing lower (30-35 mmHg) than physiological (60-80 mmHg) perfusion pressures during Langendorff.
