Latex Bead Phagocytosis

Latex bead phagocytosis is a laboratory assay that measures how immune cells engulf synthetic polymer beads, providing a model for studying cellular ingestion and innate immune function. During the process, phagocytic cells such as macrophages recognize bead surfaces, extend actin-driven pseudopods around them, and enclose them within phagosomes that can mature through intracellular processing pathways. Researchers commonly quantify bead uptake by microscopy or flow cytometry to compare phagocytic activity under different conditions, including exposure to immune signals or potential treatments. The assay supports investigations of cell biology, host defense, inflammation, and immune-cell dysfunction while offering a controlled alternative to using biological particles.

Latex agglutination testing is a simple, rapid and inexpensive method for serotyping Streptococcus pneumoniae, and has also been widely applied in diagnostic microbiology. This manuscript describes the in-house production of latex agglutination reagents, quality control procedures and the application of this technique to pneumococcal serotyping.

An In Vitro Assay to Measure Antibody-Dependent Cellular Phagocytosis by Phagocytes

2025

This video demonstrates an assay to measure the antibody-dependent cellular phagocytosis (ADCP) by breast milk phagocytes. Fluorescently labeled microspheres conjugated to an antigen-antibody complex, are combined with a cell suspension isolated from breast milk to induce antibody-dependent cellular phagocytosis by the phagocytic leukocytes. Upon labeling the cells with antibodies against common leukocyte antigen (CD45), flow cytometry is used to measure the fluorescence intensity of...

Cells pull particles inward and engulf them in spherical vesicles in an energy-requiring process called endocytosis. Phagocytosis (“cellular eating”) is one of three major types of endocytosis. Cells use phagocytosis to take in large objects—such as other cells (or their debris), bacteria, and even viruses. The objective of phagocytosis is often destruction. Cells use phagocytosis to eliminate unwelcome visitors, like pathogens (e.g., viruses and bacteria). It is perhaps unsurprising, that many...
