Map-based Cloning

Map-based cloning is a genetic strategy for identifying a gene responsible for a trait by locating it relative to known molecular markers on a chromosome. Researchers first use linkage analysis to associate the trait with markers, then narrow the genomic interval through recombination mapping and progressively examine overlapping DNA fragments, a process often called chromosome walking. Candidate genes within the refined region are sequenced and tested for mutations or functional evidence. In biology, map-based cloning helps uncover genes underlying inherited traits, disease phenotypes, and agriculturally important characteristics, providing insights into gene function and supporting molecular breeding and genetic research.
