Melanophore Stripes

Melanophore stripes are dark, patterned bands produced by the distribution of melanin-containing pigment cells, especially in zebrafish and other vertebrates. Their formation depends on coordinated melanophore migration, proliferation, and interactions with neighboring chromatophores, including xanthophores and iridophores; changes in cell position and pigment dispersion alter stripe width and contrast. Because these patterns develop through visible, tissue-level organization, they provide a useful system for studying cell communication, developmental patterning, and regeneration. Research on melanophore stripes also helps explain how genetic changes affect pigmentation, supports investigations of pigment-cell disorders, and informs broader studies of biological pattern formation.

Highly Resolved Intravital Striped-illumination Microscopy of Germinal Centers

High-resolution intravital imaging with enhanced contrast up to 120 µm depth in lymph nodes of adult mice is achieved by spatially modulating the excitation pattern of a multi-focal two-photon microscope. In 100 µm depth we measured resolutions of 487 nm (lateral) and 551 nm (axial), thus circumventing scattering and diffraction limits.

Axon guidance molecules regulate neuronal migration and targeted growth-cone navigation. We present a powerful method, the stripe assay, to assess the ability of guidance molecules to attract or repulse neurons. In this protocol, we demonstrate the stripe assay by showing FLRT2's ability to repel cultured hippocampal neurons.
