Methylene Blue

Methylene blue is a synthetic thiazine dye and redox-active compound used in biology as a cellular stain, chemical indicator, and experimental reagent. Its oxidized form appears blue, while reduction converts it to colorless leucomethylene blue; as a positively charged dye, it also associates with negatively charged cellular components, improving contrast in microscopy. These properties support visualization of cell and tissue structures, including bacterial cells, and enable assays that monitor reducing activity or cellular metabolism. Methylene blue therefore links chemical reactivity with biological observation, making it useful in microscopy, teaching laboratories, physiology, and research on cellular function.

In this video, we describe the methylene blue staining procedure to quantify the metastatic burden by counting stained colonies obtained by culturing cancer cells harvested from distant organs of a mouse model following injection.

The Application of 1% Methylene Blue Dye As a Single Technique in Breast Cancer Sentinel Node Biopsy

In Indonesia, sentinel node biopsy (SNB) is not routinely performed for breast cancer surgery because of the limitation to provide radioisotope tracer and isosulfan or patent blue dye (PBD). To overcome this obstacle, we applied 1% methylene blue dye (MBD) as a single agent to map the sentinel nodes (SNs).

The preparation and exfoliation of CaCuSi4O10 and BaCuSi4O10 are described. Upon stirring in hot water, CaCuSi4O10 spontaneously exfoliates into monolayers, whereas BaCuSi4O10 requires ultrasonication in organic solvents. Near infrared (NIR) imaging illustrates the NIR emission properties of these materials, and aqueous dispersions of these nanomaterials are useful for solution processing.
