Microscope Setup

Microscope setup is the process of arranging and adjusting a microscope for clear, accurate observation of biological specimens. It aligns the light source, condenser, stage, objectives, and eyepiece so illumination passes through the sample and the lenses produce a focused, magnified image; coarse and fine focus controls then refine image detail. Proper setup improves contrast, resolution, and ease of viewing while reducing artifacts and protecting both the specimen and instrument. In biology, these practices support routine examination of cells, tissues, microorganisms, and prepared slides, providing a reliable foundation for microscopy-based teaching, identification, and research.

A fast and inexpensive method for the behavioral determination of hearing parameters like hearing thresholds, hearing impairments or phantom perceptions (subjective tinnitus) is described. It uses pre-pulse inhibition of the acoustic startle response and can be easily implemented in a personal computer using a programmable AD/DA-converter and a piezo sensor.

Multiplexed ion beam imaging (MIBI) is often used to image tissue microarrays and tiled, contiguous tissue areas, but current software for setting up these experiments is cumbersome. The tile/SED/array interface is an intuitive, interactive graphical tool developed to dramatically simplify and accelerate MIBI run setup.

Keeping the microscope optics clean is important for high-quality imaging. Dust, fingerprints, excess immersion oil, or mounting medium on or in a microscope causes reduction in contrast and resolution. DIC is especially sensitive to contamination and scratches on the lens surfaces. This protocol details the procedure for keeping the microscope clean.

Major Components of the Light Microscope

2008

The light microscope is a basic tool for the cell biologist, who should have a thorough understanding of how it works, how it should be aligned for different applications, and how it should be maintained as required to obtain maximum image-forming capacity and resolution. The components of the microscope are described in detail here.
