Skeletal Muscle

Skeletal muscle is a type of striated muscle tissue that attaches to bones and produces voluntary movement, posture, and heat, making it essential to locomotion and whole-body function. When a motor neuron stimulates a muscle fiber, calcium ions are released from the sarcoplasmic reticulum and bind to regulatory proteins, allowing myosin heads to interact with actin and generate force through repeated cross-bridge cycling. Skeletal muscle also adapts to exercise, disuse, injury, and changes in neural input, making it an important model for studying cellular organization, energy metabolism, regeneration, and neuromuscular disease. Understanding its structure and function supports research in physiology, rehabilitation, sports science, and medicine.

Mitochondrial Isolation from Skeletal Muscle

This protocol describes a procedure to study the respiration of mitochondria isolated from skeletal muscles. This method was adapted from Scorrano et al. (2007). The mitochondrial isolation procedure requires about 2 hours. The mitochondrial respiration can be completed in about 1 hour.

The micro-dissected explants technique is a robust and reliable method for isolating proliferative skeletal muscle cells from juvenile, adult or embryonic muscles as a source of skeletal muscle stem cells. Uniquely, these cells have been clonally derived to produce skeletal muscle stem cell lines used for in vivo transplantation.

A straight-forward set of methods to isolate and determine the identity of the most abundant proteins expressed in skeletal muscle. About 800 spots are discerned on a two-dimensional gel from 10 mg muscle; this allows for the determination of gender-specific protein expression. These methods will give equivalent results in most tissues.
