Midgut Isolation

Midgut isolation is a dissection-based method for separating the midgut, the central region of an animal’s digestive tract, from surrounding tissues for focused biological analysis. The procedure typically involves exposing the digestive system, identifying the midgut by its anatomical position and structure, and carefully removing it while minimizing damage and contamination. Isolated midgut tissue supports studies of digestion, nutrient absorption, epithelial organization, host–microbe interactions, and responses to chemicals or pathogens. By providing a defined tissue preparation, the method helps researchers examine gut physiology and molecular changes that may be difficult to distinguish in whole-organism samples.

Understanding the endogenous molecular changes in adult stem cells during aging requires isolating the cells of interest. The method described here presents a simple and robust approach to enrich for and isolate Drosophila intestinal stem cells and the enteroblast progenitor cells by FACS at any time point during aging.
