Mitochondria Morphology

Mitochondrial morphology describes the size, shape, distribution, and internal organization of mitochondria, membrane-bound organelles that support cellular energy production and signaling. Their appearance reflects a dynamic balance between fusion, which joins mitochondria and promotes content mixing, and fission, which divides them and helps distribute organelles or remove damaged regions through mitophagy; cristae architecture within the inner membrane also influences respiratory function. Examining these features with fluorescence microscopy, electron microscopy, or live-cell imaging helps researchers connect mitochondrial structure with metabolism, cell division, stress responses, and disease. Morphological changes can therefore indicate cellular dysfunction and guide biological investigation.

Multi-parameter Measurement of the Permeability Transition Pore Opening in Isolated Mouse Heart Mitochondria

A spectrofluorometric protocol for the measurement of the mitochondrial permeability transition pore opening in isolated mouse heart mitochondria is presented here. The assay involves the simultaneous measurement of mitochondria Ca2+ handling, mitochondrial membrane potential and mitochondrial volume. The procedure for obtaining high-quality and functional heart mitochondria is also described.

Purification of Mitochondria from Yeast Cells

We describe a rapid and effective method for purification of mitochondria from the yeast Saccharomyces cerevisiae. This method enables the high-yield isolation of pure mitochondria that are essentially free of contamination by other organelles and retain their structural and functional integrity after their purification.
