Calcium Transients

Calcium transients are rapid, temporary changes in intracellular calcium ion (Ca2+) concentration that act as signals in living cells. They arise when stimuli open calcium-permeable channels in the plasma membrane or trigger Ca2+ release from intracellular stores such as the endoplasmic or sarcoplasmic reticulum; pumps, exchangers, and buffering proteins then restore baseline levels. These localized or cell-wide signals regulate processes including neurotransmitter release, muscle contraction, secretion, metabolism, and gene expression. In biology, calcium imaging and fluorescent indicators allow researchers to measure transient timing, amplitude, and spatial patterns, providing insight into cell communication, development, physiology, and disease mechanisms.

Here, we describe a method for loading a calcium-sensitive dye through the frog nerve stump into the nerve endings. We also present a protocol for the recording and analysis of fast calcium transients in the peripheral nerve endings.
