Mrna Microinjection

mRNA microinjection is a laboratory technique that delivers messenger RNA directly into a cell, allowing researchers to produce a chosen protein without introducing DNA into the genome. Using a fine glass needle, an mRNA solution is injected into the cytoplasm or, in some systems, a fertilized egg, where cellular ribosomes translate the transcript into protein before the RNA is naturally degraded. In biology, this approach enables transient gene expression in embryos, oocytes, and cultured cells. It supports studies of gene function, early development, cell signaling, and protein activity while providing precise control over expression timing and dose.

Zebrafish Microinjection Techniques

2023

One of the major advantages to working with zebrafish (Danio rerio) is that their genetics can be easily manipulated by microinjection of early stage embryos. Using this technique, solutions containing genetic material or knockdown constructs are delivered into the blastomeres: the embryonic cells sitting atop the yolk of the newly fertilized egg. Delivery into the cytoplasm is achieved either through direct injection into the blastomere, or via natural cytoplasmic movements that occur after a...

Regulated mRNA Transport

2020

In eukaryotes, transcription and translation are compartmentalized; an mRNA is first synthesized in the nucleus and then selectively transported to the cytoplasm for protein synthesis. Before transport, a pre-mRNA undergoes several steps of post-transcriptional modifications including splicing, 5' capping, and the addition of a poly-adenine tail. Various proteins bind to the pre-mRNA during these modifications. The mRNA transport takes place with the help of multiple proteins playing specific...
