Bone Marrow Engraftment

Bone marrow engraftment is the process by which transplanted hematopoietic stem cells establish themselves in the recipient’s bone marrow and restore blood-cell production. After conditioning therapy creates space and suppresses existing marrow activity, infused stem cells migrate to marrow niches, where they survive, self-renew, and differentiate into red blood cells, white blood cells, and platelets. Engraftment is a critical milestone in bone marrow and hematopoietic stem-cell transplantation because it indicates recovery of hematopoiesis and reduced dependence on transfusions. Monitoring cell counts helps assess graft function, while delayed or failed engraftment can increase the risk of infection, bleeding, and other complications.

Determining donor cell engraftment presents a challenge in mouse bone marrow transplant models that lack well-defined phenotypical markers. We described a methodology to quantify male donor cell engraftment in female transplant recipient mice. This method can be used in all mouse strains for the study of HSC functions.

The recent explosion of knowledge pertaining to evolutionary, genetic, and biochemical aspects of the interaction between macrophages and microbes has renewed scientific attention to macrophages. Murine Hind Limb Long Bone Dissection and Bone Marrow Isolation

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.
