Nanometer Localization

Nanometer localization is the determination of a molecule’s or structure’s position with nanometer-scale precision, enabling researchers to resolve biological organization below the diffraction limit of light microscopy. The method typically records light from an individual fluorescent emitter and fits its diffraction-limited point-spread function to estimate the emitter’s center, with precision improved by collecting more photons and reducing background noise. Applied in localization microscopy, this approach maps proteins, nucleic acids, and cellular structures over successive imaging cycles. It supports studies of molecular organization, trafficking, and interactions, providing quantitative spatial information for cell biology and biophysics.

We present a method for achieving sub-nanometer resolution images with amplitude-modulation (tapping mode) atomic force microscopy in liquid. The method is demonstrated on commercial atomic force microscopes. We explain the rationale behind our choices of parameters and suggest strategies for resolution optimization.

Nano-fEM: Protein Localization Using Photo-activated Localization Microscopy and Electron Microscopy

We describe a method to localize fluorescently tagged proteins in electron micrographs. Fluorescence is first localized using photo-activated localization microscopy on ultrathin sections. These images are then aligned to electron micrographs of the same section.
