Olfactory Receptors

Olfactory receptors are specialized proteins that detect odorant molecules and convert chemical information into the neural signals underlying the sense of smell. Located primarily on the cilia of olfactory sensory neurons, these G protein-coupled receptors activate the G protein Golf when an odorant binds, stimulating adenylyl cyclase to increase cyclic AMP and open ion channels that depolarize the neuron. The resulting receptor activity creates patterns interpreted by the brain as distinct odors. Studying olfactory receptors clarifies how organisms recognize environmental chemicals, supports research on sensory coding and behavior, and informs the development of odor-detection technologies and potential biomedical applications.

Olfactory receptor activation patterns encode odor identity, but the lack of published data identifying odorant ligands for mammalian olfactory receptors hinders the development of a comprehensive model of odor coding. This protocol describes a method to facilitate high-throughput identification of olfactory receptor ligands and quantification of receptor activation.

Olfactory receptor neurons (ORNs) convert odor signals first into a receptor current that in turn triggers action potentials that are conveyed to second order neurons in the olfactory bulb. Here we describe the suction pipette technique to record simultaneously the odorant-induced receptor current and action potentials from mouse ORNs.

Cilia of olfactory sensory neurons contain proteins of the signal transduction cascade, but a detailed spatial analysis of their distribution is difficult in cryosections. We describe here an optimized approach for whole mount labeling and en face visualization of ciliary proteins.
