Origin Of Transfer

The origin of transfer, or oriT, is a specific DNA sequence where plasmid transfer begins during bacterial conjugation, making it central to the movement of genetic information between cells. A relaxase enzyme recognizes oriT, introduces a site-specific nick at the nic site, and remains attached to the transferred DNA strand as conjugation machinery moves it into a recipient cell; complementary-strand synthesis then restores a double-stranded plasmid. Studying oriT helps researchers characterize mobilizable plasmids, investigate horizontal gene transfer, and understand how antibiotic-resistance and other adaptive genes spread through bacterial populations.

A method of constructing a phylogenetic tree based on sequence homology of SWEETs from eukaryotes and SemiSWEETs from prokaryotes is described. Phylogenetic analysis is a useful tool for explaining the evolutionary relatedness between homologous proteins or genes from different organism groups.
