Oxa Protein Assembly

Oxa protein assembly is the membrane-insertion and maturation process that places selected proteins into the inner membrane of mitochondria and related organelles, where they support energy conversion. The Oxa translocase recognizes newly synthesized polypeptides, often as they emerge from mitochondrial ribosomes, moves hydrophobic segments toward the membrane, and releases them laterally into the lipid bilayer for folding and association with partner subunits. This pathway is essential for building respiratory-chain complexes and maintaining membrane function. Studying Oxa-mediated assembly clarifies how organelle-encoded proteins become functional and helps explain defects in mitochondrial biogenesis, protein homeostasis, and cellular energy production.

Protein Complex Assembly

2020

Proteins can form homomeric complexes with another unit of the same protein or heteromeric complexes with different types. Most protein complexes self-assemble spontaneously via ordered pathways, while some proteins need assembly factors that guide their proper assembly. Despite the crowded intracellular environment, proteins usually interact with their correct partners and form functional complexes. Many viruses self-assemble into a fully functional unit using the infected host cell to...

Source: Olety, B., et. al., Visualization of HIV-1 Gag Binding to Giant Unilamellar Vesicle (GUV) Membranes. J. Vis. Exp. (2016)This study demonstrates a fluorescence-based assay to observe the membrane-binding behavior of human immunodeficiency virus type-1 structural protein. Using synthetic giant unilamellar vesicles as model membranes, the assay visualizes protein recruitment and assembly, mimicking early steps of viral particle formation.
