Pancreatic Secretions

Pancreatic secretions are digestive fluids released by the exocrine pancreas into the duodenum, where they support the breakdown and absorption of nutrients. Acinar cells produce enzymes such as amylase, lipase, and protease precursors, while duct cells add bicarbonate-rich fluid that neutralizes acidic chyme from the stomach; hormones including secretin and cholecystokinin coordinate this response. Studying pancreatic secretions helps explain digestive physiology, nutrient processing, and regulation of the gastrointestinal system. Their composition and flow also provide important biological context for disorders such as pancreatitis, exocrine pancreatic insufficiency, and impaired digestion.

Pancreatic Juice and Secretion

Pancreatic juice is a clear fluid produced by the pancreas, containing water, salts, sodium bicarbonate, and enzymes vital for digestion in the small intestine. It helps break down large molecules, facilitating nutrient absorption. When acidic chyme from the stomach enters the duodenum, it triggers the release of secretin, a hormone that prompts pancreatic juice secretion. After a fatty meal, cholecystokinin, another hormone, stimulates gallbladder contraction and enhances enzyme-rich...

Glucose Homeostasis: Pancreatic Islets and Insulin Secretion

2024

The pancreatic islets comprising only 1%-2% of the volume are highly vascularized and innervated mini-organs. They contain five endocrine cell types, including β cells that secrete insulin, which is synthesized as a single polypeptide chain, preproinsulin, processed to proinsulin, and finally to insulin and C-peptide. This process is complex and regulated, involving the Golgi complex, the endoplasmic reticulum, and the secretory granules of the β cell. Insulin and C-peptide are co-secreted in...

Transcellular protein interactions are important determinants of pancreatic beta-cell function. Detailed here is a method—adapted from a coculture model of synaptogenesis—for investigating how specific transmembrane proteins influence insulin secretion. Transfected HEK293 cells express proteins of interest; beta cells do not need to be transfected or otherwise directly perturbed.

2023

This video describes the protocol for grafting human pancreatic islets into the anterior chamber of the mouse’s eye to monitor revascularization of human islet grafts in vivo. It also helps to understand the contribution of recipient versus donor cells in promoting the encapsulation and vascularization of the graft.
