Plant Proteins

Plant proteins are amino acid-based molecules produced by plants that perform structural, enzymatic, transport, storage, and signaling functions, while also contributing to human nutrition. Their synthesis begins when genes are transcribed into messenger RNA and ribosomes translate that information into polypeptide chains, which fold into functional three-dimensional structures; amino acid composition and cellular conditions influence their properties. In biology, studying plant proteins helps explain growth, metabolism, stress responses, and seed development. Extracted proteins from legumes, grains, and other crops also support nutritional research, food formulation, and the development of sustainable alternatives to animal-derived protein.

Analysis of Protein Import into Chloroplasts Isolated from Stressed Plants

Here we describe a new method to study protein import into isolated chloroplasts under stress. The method is rapid and straightforward, and can be applied to study the consequences of different stress conditions for chloroplast protein import, and the corresponding regulatory mechanisms.

Soluble Protein Assay to Quantify Soluble Protein Content in Plant Tissues

This video describes a colorimetric method to quantify the soluble protein content in plant tissues. The method uses Coomassie Brilliant Blue G-250 dye, which interacts with the soluble protein to form a blue-colored stable protein-dye complex, proportional to the soluble protein content in the tissue.

Immunolocalization of Arabinogalactan Proteins and Pectins in Plant Tissue

This video demonstrates an immunostaining technique to localize arabinogalactan proteins (AGPs) and pectins in plant tissues. Fixed and resin-embedded plant tissue sections are stained with AGP- and pectin-specific primary and fluorescent secondary antibodies and a cellulose-binding fluorescent dye to reveal their tissue-specific colocalization in the cell wall.

Identification of Post-translational Modifications of Plant Protein Complexes

We describe here a protocol for the purification and characterization of plant protein complexes. We demonstrate that by immunoprecipitating a single protein within a complex, so we can identify its post-translational modifications and its interacting partners.

Plants offer a novel system for the production of pharmaceutical proteins on a commercial scale that is more scalable, cost-efficient and safe than current expression paradigms. In this study, we report a simple and convenient, yet scalable approach to introduce target-gene containing Agrobacterium tumefaciens into plants for protein transient expression.
