Plasmid Cloning

Plasmid cloning is a molecular biology method for inserting a DNA sequence into a circular, self-replicating plasmid so the sequence can be copied, analyzed, or expressed in host cells. The process typically joins an insert with a plasmid vector using restriction enzymes and DNA ligase or a compatible assembly method, then introduces the recombinant plasmid into bacteria by transformation. Antibiotic selection identifies cells carrying the vector, while colony screening and DNA sequencing confirm the intended construct. Plasmid cloning supports gene function studies, protein production, genetic engineering, and the development of research tools across biology.
