Macrophage Signaling

Macrophage signaling is the network of molecular communication processes that enables macrophages to detect environmental cues and coordinate immune and tissue responses. Signals from pattern-recognition receptors, cytokine receptors, and other surface sensors activate intracellular pathways such as NF-κB and JAK-STAT, which alter gene transcription and regulate cytokine release, phagocytosis, and cellular polarization. These pathways help macrophages respond to infection, control inflammation, and support tissue repair, while dysregulated signaling can contribute to chronic inflammatory disease, cancer, and metabolic disorders. Studying macrophage signaling therefore provides a framework for understanding innate immunity and identifying potential therapeutic targets.

Investigation of Macrophage Polarization Using Bone Marrow Derived Macrophages

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.

This video shows the co-culturing of murine dorsal root ganglion neurons and macrophages. Neurons are seeded on a coated plate, while macrophages are collected from an euthanized mouse, treated, and added to the culture insert. Treatment with db-cAMP activates neurons and prompts macrophages to adopt a pro-regenerative phenotype to support neuronal outgrowth.

A protocol is presented for cell culture of macrophage colony-stimulating factor (M-CSF) differentiated human monocyte-derived macrophages. The protocol utilizes cryopreservation of monocytes coupled with their bulk differentiation into macrophages. Then harvested macrophages can then be seeded into culture wells at required cell densities for carrying out experiments.

Macrophages have long been recognized as a critical component of the innate and adaptive immune responses. The recent explosion of knowledge pertaining to evolutionary, genetic, and biochemical aspects of the interaction between macrophages and microbes has renewed scientific attention to macrophages.
