Plaster Charcoal Substrate

Plaster charcoal substrate is a porous, moisture-retaining culture surface made by combining plaster with charcoal, providing a stable environment for biological growth and observation. As the plaster hardens, it forms a firm matrix, while charcoal contributes an uneven, absorbent structure that helps retain moisture and creates surface spaces for organisms or cultures. In biology, this substrate supports laboratory rearing and maintenance of organisms that require humid, structured conditions, enabling researchers to monitor growth, development, survival, or behavior under controlled conditions. Its reproducible composition and accessible materials make it useful for teaching laboratories and experimental systems involving small organisms or microbial cultures.

The charcoal agar resazurin assay (CARA) is a semi-quantitative, medium-throughput method to assess activity of test agents against mycobacteria that are replicating, non-replicating, or both. The CARA permits rapid evaluation of time- and concentration-dependent activity and identifies parameters to pursue by colony forming unit (CFU) assays.

Protein phosphorylation is a central feature of how cells interpret and respond to information in their extracellular milieu. Here, we present a high throughput screening protocol using kinases purified from mammalian cells to rapidly identify kinases that phosphorylate a substrate(s) of interest.

Substrate Oxidation Assay in Cultured Cells: An In Vitro Assay to Quantify Substrate Oxidation in Cells by Measuring Radioactive Signals from Trapped CO2

This video demonstrates a quantification technique for CO2 released during substrate oxidation using 14C-radiolabeled substrates. The released 14CO2 is trapped in an alkaline solution and quantified by a scintillation counter. The oxidation of different substrates varies between tissues and reflects the pathophysiological condition of the tissue.
