Polyq Aggregation

PolyQ aggregation is the abnormal self-assembly of proteins containing expanded stretches of glutamine residues, a process linked to several inherited neurodegenerative disorders. Excessive polyglutamine sequences can destabilize protein structure and promote intermolecular hydrogen bonding, producing soluble oligomers and, in many cases, insoluble amyloid-like fibrils and intracellular inclusions. Studying this aggregation process helps explain how genetic repeat expansions disrupt neuronal function and survival in conditions such as Huntington’s disease and several spinocerebellar ataxias. Experimental models of polyQ aggregation also support research into protein quality control, cellular clearance pathways, disease biomarkers, and potential strategies for preventing or reducing toxic protein assemblies.

In this video, we describe a method to evaluate the neuroprotective effect of a test compound, astragalan, on neuronal survival in Caenorhabditis elegans from polyQ-mediated neurotoxicity. The assay uses Caenorhabditis elegans expressing the abnormally expanded polyQ repeats fused to GFP in their ASH neurons to measure neuronal survival following test compound treatment.
