Pre-initiation Complex

The pre-initiation complex is a multi-protein assembly that forms at a gene promoter to position RNA polymerase II for accurate transcription initiation. In eukaryotic cells, general transcription factors bind promoter DNA, recruit and stabilize RNA polymerase II, and assemble factors including TFIID, TFIIA, TFIIB, TFIIF, TFIIE, and TFIIH; TFIIH then unwinds promoter DNA and phosphorylates the polymerase C-terminal domain to enable promoter escape. Studying this complex clarifies how regulatory signals control gene expression and how promoter architecture influences transcription. Its components are important in research on development, cellular differentiation, disease-associated transcription defects, and mechanisms targeted by experimental therapeutics.
