Rat Hippocampus Isolation

Rat hippocampus isolation is a neuroanatomical dissection technique used to separate the hippocampus from the rat brain for structural, cellular, or molecular analysis. The procedure relies on recognizing hippocampal anatomy within the medial temporal region and carefully removing the tissue from surrounding cortical and subcortical structures, often while preserving its integrity for downstream processing. Isolated hippocampal tissue can support studies of learning and memory, synaptic plasticity, neurogenesis, neuronal injury, and disease mechanisms. Depending on the experimental goal, researchers may use the sample for histology, gene or protein analysis, electrophysiology, or preparation of primary neuronal cultures.

This video demonstrates a method for preparing organotypic hippocampal slice cultures from the brain of a rat pup. It outlines the steps involved in the dissection of the hippocampus, slicing, and culture conditions required to promote cell viability and growth in the hippocampal slices, ultimately generating organotypic slice cultures.

The mechanism underlying the therapeutic effects of Deep Brain Stimulation (DBS) surgery needs investigation. The methods presented in this manuscript describe an experimental approach to examine the cellular events triggered by DBS by analyzing the gene expression profile of candidate genes that can facilitate neurogenesis post DBS surgery.

Controlled Infusion of Aβ Peptide-Specific Antibodies and Aβ Oligomers into the Rat Hippocampus

2025

This video demonstrates a technique for the continuous infusion of amyloid-beta (Aβ) peptide-specific antibodies into the hippocampus of a rat. The study employs implanting a bilateral cannula system for a controlled infusion of Aβ peptide-specific antibodies coupled with the infusion of soluble Aβ oligomers (Aβo) into the rat hippocampus to study the interactions between the oligomers and the antibodies.
