Respiratory Chain Complexes

Respiratory chain complexes are protein assemblies in the inner mitochondrial membrane that transfer electrons and support cellular energy production through oxidative phosphorylation. Complexes I, II, III, and IV pass electrons from NADH and FADH2 to oxygen, using the released energy to pump protons across the membrane and create an electrochemical gradient. ATP synthase then uses this gradient to convert ADP and inorganic phosphate into ATP, while oxygen is reduced to water at the end of the chain. Studying these complexes helps explain metabolism, mitochondrial disease, reactive oxygen species, and how cells adapt energy production to physiological demands.

In this video, we demonstrate an electrophoretic technique, called blue native PAGE, for the analysis of mitochondrial respiratory protein complexes in their natively folded and functionally active state. The anionic Coomassie blue dye causes the protein complexes to become negatively charged, allowing separation based on size and structure.

This enables the characterization of conserved genes' novel functions in the mitochondrial respiratory chain.
