This article describes a method for analyzing mitochondrial complexes of the respiratory chain using blue native gel electrophoresis. The technique allows for the preservation of the native structure and function of the complexes while enabling their separation based on size.
Blue native PAGE enables preservation of mitochondrial respiratory chain complexes in their native, functional state for size-based separation. This supports target validation in mitochondrial dysfunction pathways by maintaining complex integrity during analysis. The technique provides predictive confidence in early discovery by resolving intact complexes for mechanistic de-risking.
The method integrates into the discovery continuum from early target validation through lead identification to preclinical work by enabling analysis of intact respiratory chain complexes.
