Salt Gradient Elution

Salt gradient elution is a chromatography technique that separates charged molecules by gradually increasing the salt concentration in the mobile phase. In ion-exchange chromatography, rising ionic strength introduces competing ions that weaken electrostatic interactions between proteins and oppositely charged resin sites, causing weakly bound molecules to elute before strongly bound ones. The resulting elution profile helps researchers separate, identify, and purify proteins, nucleic acids, and other biomolecules based on charge. By controlling the gradient slope, starting and ending salt concentrations, and flow conditions, scientists can improve resolution and recover biologically active compounds for biochemical analysis, structural studies, and biotechnology applications.
