Sarcoplasmic Reticulum

The sarcoplasmic reticulum is a specialized form of endoplasmic reticulum in muscle cells that stores and regulates calcium ions, making it essential for contraction and relaxation. When an action potential travels along the sarcolemma and into the transverse tubules, it activates calcium-release channels called ryanodine receptors, allowing calcium to enter the cytoplasm and initiate actin-myosin interaction; afterward, SERCA pumps return calcium to the reticulum. This tightly controlled cycle supports skeletal, cardiac, and smooth muscle function. Studying the sarcoplasmic reticulum helps explain excitation-contraction coupling and provides insight into muscle disorders, cardiovascular disease, and potential therapeutic targets.

We describe the imaging approaches we use to investigate the distribution and mobility of the transfected fluorescent proteins resident in the endoplasmic reticulum (ER) by means of the confocal imaging of living cells. We also ultrastructurally analyze the effect of their expression on the architecture of this subcellular compartment.
