Sgrna Complex

The sgRNA complex is a ribonucleoprotein assembly in which a single-guide RNA (sgRNA) associates with a CRISPR-associated nuclease, most commonly Cas9, to direct sequence-specific DNA targeting. The sgRNA combines a guide sequence that base-pairs with a complementary DNA region and a scaffold that binds and activates the nuclease; when a compatible protospacer-adjacent motif is present, the complex can form an RNA-DNA hybrid and promote localized DNA cleavage. In biology, sgRNA complexes support targeted genome editing, gene-function studies, and regulation of gene expression, while their design and delivery influence targeting efficiency, specificity, and experimental outcomes.

This video demonstrates a technique for nanoblade-based delivery of Cas9-sgRNA ribonucleoprotein complex inside target cells for genome editing. Nanoblades are virus-like particles devoid of the ability to multiply and infect neighboring cells, therefore are very useful for rapid and dose-dependent transportation of bio- and nanomaterials.

We have developed a simple and inexpensive protocol to load Cas9/single-guide RNA (sgRNA) ribonucleoprotein complexes within virus-like particles. These particles, called "Nanoblades", allow efficient delivery of the Cas9/sgRNA complex in immortalized and primary cells as well as in vivo.
