Soil Health Activity

A Soil Health Activity is a structured investigation of how well soil supports plant growth and living organisms, making it a practical way to connect biology with environmental conditions. Students examine soil properties such as texture, moisture, organic matter, pH, and visible biological activity, then compare observations or measurements among samples to evaluate soil function. These activities show how physical, chemical, and biological factors interact within an ecosystem. High-throughput Fluorometric Measurement of Potential Soil Extracellular Enzyme Activities

To measure potential rates of soil extracellular enzyme activities, synthetic substrates that are bound to a fluorescent dye are added to soil samples. Enzyme activity is measured as the fluorescent dye is released from the substrate by an enzyme-catalyzed reaction, where higher fluorescence indicates more substrate degradation.

Microplate based procedures are described for the colorimetric or fluorometric analysis of extracellular enzyme activity. These procedures allow for the rapid assay of such activity in large numbers of environmental samples within a manageable time frame.
