Split Biotin Ligase

Split biotin ligase is a proximity-labeling tool made by dividing a biotin-protein ligase into inactive fragments that report when two molecular partners come together. When the fragments are recruited to the same cellular site, they reassemble into an active enzyme, convert biotin into a reactive intermediate, and covalently attach biotin to nearby proteins. Researchers can then isolate labeled proteins with streptavidin and identify interaction partners or local protein neighborhoods by mass spectrometry or imaging. In biology, this approach supports studies of protein-protein interactions, organelle contacts, and signaling dynamics while reducing labeling away from the site of interest.
