Synthetic Calcium Indicators

Synthetic calcium indicators are small fluorescent dyes that report changes in intracellular calcium concentration, making rapid cell-signaling events visible in living cells. They typically contain a calcium-binding chelator linked to a fluorophore; when calcium binds, the dye’s fluorescence intensity, wavelength, or excitation properties change, allowing optical measurement of calcium dynamics with microscopy. Researchers load these indicators into cells using membrane-permeant ester forms, microinjection, or related delivery methods, then track signals associated with neuronal activity, muscle contraction, secretion, and intracellular signaling. Their tunable colors, affinities, and kinetics support quantitative comparisons across biological experiments, although dye loading and buffering can influence cell physiology and measurement accuracy.

Loading Drosophila Nerve Terminals with Calcium Indicators

Calcium is a ubiquitous messenger in the nervous system, essential for triggering neurotransmitter release and changes in synaptic strength. Here we demonstrate a technique for loading Ca2+-indicators into Drosophila nerve terminals. We also demonstrate fabrication of the required apparatus and emphasize points critical for the technique's success.
