Thermogenic Adipocytes

Thermogenic adipocytes are specialized fat cells that generate heat rather than storing energy, making them important regulators of body temperature and energy balance. In brown and beige adipocytes, sympathetic stimulation activates mitochondrial uncoupling protein 1 (UCP1), which dissipates the proton gradient produced during respiration as heat instead of using it to synthesize ATP. Their activity contributes to non-shivering thermogenesis, particularly during cold exposure, and is influenced by developmental and environmental signals. Studying these cells helps clarify metabolic regulation and may support research into obesity, diabetes, and therapies that increase energy expenditure.

Adipose tissue (AT) can influence whole body homeostasis, therefore understanding the molecular mechanisms of adipocyte differentiation and function is of importance. We provide a protocol for gaining new insights into these processes by analyzing adipocyte homeostasis, differentiation and hypoxia exposure as a model for induced adipocyte apoptosis.

This video demonstrates the differentiation of bone marrow stromal cells (BMSCs) into adipocytes. In the two-step process, in the presence of specific growth factors and hormones, undifferentiated BMSCs first differentiate into preadipocytes. The second step involving treatment with glucose and insulin leads to the formation of mature adipocytes with multiple lipid droplets.

Mature adipocytes may represent an abundant source of stem cells through dedifferentiation, which leads to a homogenous population of fibroblast-like cells. Collagenase digestion is used to isolate mature adipocytes from human fat. The goal of our protocol is to obtain multipotent, dedifferentiated fat cells from human mature adipocytes.
