Thp-1 Reporter Cells

THP-1 reporter cells are engineered human monocytic cells that produce a measurable signal when specific immune signaling pathways become activated, making them useful tools in cell biology and immunology. Typically, a reporter gene such as luciferase or a fluorescent protein is placed under the control of a response-sensitive promoter; stimulation then triggers pathway-dependent gene expression and generates a quantifiable signal. These cells can model innate immune responses, inflammatory signaling, and pathogen or drug interactions in a controlled laboratory system. Their standardized readouts support pathway analysis, assay development, toxicity testing, and screening for compounds that regulate immune activation.

This protocol is based on LPS and ATP-induced death of PMA-differentiated THP-1 macrophages. We use flow cytometry to analyze Annexin V and 7-AAD double staining to detect cell death, using the whole cell and employing scanning electron microscopy to observe the cell membrane morphology.
