Thylakoid Lumen

The thylakoid lumen is the aqueous compartment enclosed by thylakoid membranes in chloroplasts and cyanobacteria, where essential steps of photosynthetic energy conversion occur. During the light-dependent reactions, water splitting and electron transport move protons into the lumen, creating an electrochemical gradient across the membrane; ATP synthase uses this gradient to produce ATP as protons return to the stroma. The lumen also contains proteins that support photosystem function, electron transfer, and regulation of photosynthesis. Studying its composition and pH helps explain how plants and microorganisms convert light energy into chemical energy and adapt to changing environmental conditions.

After the ingestion of food supplemented with a pH indicator dye, the acidification of the intestinal lumen results in a change in the color of the dye.
