Time Point Sampling

Time point sampling is a study design and data-collection approach that measures biological systems at defined moments to track change over time. Researchers collect specimens, observations, or measurements at scheduled intervals, using consistent procedures and conditions so that differences between time points reflect biological processes rather than sampling variation. In biology, this approach can reveal patterns in development, disease progression, gene expression, cellular responses, and treatment effects. Comparing measurements across a time series helps establish temporal relationships, identify short-lived or delayed responses, and determine when key biological changes occur, supporting experimental interpretation and more precise research conclusions.

Time-lapse microscopy allows the visualization of developmental processes. Growth or drift of samples during image acquisition reduces the ability to accurately follow and measure cell movements during development. We describe the use of open source image processing software to correct for three dimensional sample drift over time.

A novel sample holder for macromolecular X-ray crystallography along with a suitable handling protocol is presented. The system allows crystal growth, crystal soaking and in situ diffraction data collection at both, ambient and cryogenic temperature without the need of any crystal manipulation or mounting.
