Trajectory Segmentation

Trajectory segmentation is a computational method for dividing a time-ordered path into biologically meaningful segments, allowing researchers to distinguish different phases of movement or behavior. It analyzes changes in spatial position and temporal dynamics, such as shifts in direction, speed, persistence, or behavioral state, to identify boundaries between segments. In biology, trajectory segmentation helps interpret animal movement, cell migration, developmental processes, and microscopy-based tracking data. By reducing complex paths to discrete behavioral or functional episodes, it supports quantitative comparisons, reveals transitions that may be hidden in average measurements, and improves models of biological dynamics.

We describe a protocol to observe and analyze cell rolling trajectories on asymmetric receptor-patterned substrates. The resulting data are useful for engineering of receptor-patterned substrates for label-free cell separation and analysis.

How neuronal networks are established in the embryonic brain is a fundamental question in developmental neurobiology. Here we combined an electroporation technique with novel genetic tools, such as Cre/Lox–plasmids and PiggyBac-mediated DNA transposition system in the avian hindbrain to label dorsal interneurons and track their axonal projections and synaptic targets at various developmental stages.

Segmentation of three-dimensional data from many imaging techniques is a major bottleneck in analysis of complex biological systems. Here, we describe the use of SuRVoS Workbench to semi-automatically segment volumetric data at various length-scales using example datasets from cryo-electron tomography, cryo soft X-ray tomography, and phase contrast X-ray tomography techniques.
