Trna Splicing

tRNA splicing is the RNA-processing pathway that removes introns from precursor transfer RNAs (pre-tRNAs) and joins their exons to produce functional molecules for protein synthesis. In the canonical pathway, a tRNA splicing endonuclease cleaves at the intron boundaries, generating separate exon fragments; a tRNA ligase then repairs and seals the backbone, while additional processing can restore mature RNA ends and modify nucleotides. This coordinated mechanism ensures accurate codon recognition and amino acid delivery during translation. Studying tRNA splicing helps explain gene expression across organisms, identify defects linked to human disease, and advance research on RNA processing and cellular evolution.

This protocol describes the preparation of a synthetic tRNA substrate for the Entamoeba histolytica DNA/tRNA methyltransferase 2 (Dnmt2) homolog Ehmeth and the measure of its methyltransferase activity. This experimental approach can be used for investigating the activity of other Dnmt2 proteins.
